The dose depends on which trial's evidence you're chasing
NMN doesn't have one settled dose the way some nutrients do — eight verified human trials used doses from 250 to 1250 mg/day, and the honest approach is understanding which dose goes with which finding, not defaulting to the biggest number on the shelf. Four of the eight trials all used 250 mg/day — but read each carefully: Yoshino 2021 (insulin sensitivity — though a published critique found the two arms started with very different liver fat, 6.3% vs 14.8%, p=0.003, which undercuts the result), the Mitsubishi-funded Igarashi 2022 (nominally significant gait-speed and grip gains), Kim 2022 (primary 5-times-sit-to-stand test: null; secondary sleep-quality benefit only), and Morifuji 2024 (primary stepping test: null; secondary walking-speed and sleep gains). That makes 250 mg both the most-studied and the most common retail dose. The two dose-ranging trials tell a different story: Liao 2021 tested 300, 600, and 1200 mg/day in runners and found aerobic-capacity gains rose with dose, and Yi 2023 (co-authors affiliated with NMN maker Abinopharm) tested 300, 600, and 900 mg/day and found 6-minute walk distance improved at all three doses, with 300 mg the weakest responder and 600-900 mg producing the longest distances. Neither trial tested doses above 1200 mg for efficacy. Safety, separately, has only been pushed to 1250 mg/day, and only for 4 weeks (Fukamizu 2022).
