B-Complex Dosage Guide: What Each B Does & the 100 mg B6 Upper Limit

The B vitamins are cofactors — they help most when they correct a deficiency, not as a top-up on a normal level. Two dosing facts matter most: vitamin B6 has a 100 mg/day Upper Limit (NIH ODS / IOM) tied to a reversible nerve neuropathy (Schaumburg 1983), and lowering homocysteine does not cut cardiovascular events despite the biomarker drop (Martí-Carvajal 2017; Lonn 2006). So dose for adequacy and a safe B6 — not for "high potency."
What each B vitamin actually does
Every B vitamin is an enzyme cofactor — a helper molecule a specific reaction can't run without. That's the key to dosing sanely: a cofactor is only rate-limiting when it's missing, so correcting a shortfall helps, but stacking more onto a normal level generally doesn't do anything extra.

The B6 Upper Limit: 100 mg/day
Vitamin B6 is the one B vitamin with a real toxicity ceiling. The tolerable Upper Intake Level is 100 mg/day for adults, set by the NIH Office of Dietary Supplements and the Institute of Medicine (an authoritative reference value, not a single study). The reason is a sensory peripheral neuropathy — tingling, numbness, and an unsteady gait — documented in people taking large B6 doses over time (Schaumburg 1983). It's usually reversible once B6 is stopped, but it's a real, avoidable harm. The trap: many "B-50" and "B-100" products put B6 at 50–100 mg per serving, so a single high-potency bottle can reach the limit by itself — and 2 of the 11 products we track sit right at 100 mg. There's no established benefit to those doses outside a diagnosed deficiency, so keeping B6 well under 100 mg/day is the cautious default. Homocysteine down, heart risk unchanged A B-complex reliably lowers homocysteine — but that is not a cardiovascular benefit. The Cochrane review found no drop in heart attack or death (Martí-Carvajal 2017), HOPE-2 found no CV benefit and a signal of more unstable angina (Lonn 2006), and VISP was null (Toole 2004). So "lower your homocysteine to protect your heart" is exactly the claim the trials refuted. Don't dose a B-complex for cardiovascular prevention.

How to dose it sensibly
- Dose for adequacy, not "potency." The benefit is correcting a shortfall; a modest, complete B-complex does that. A megadose doesn't do more — it just adds B6 risk.
- Keep B6 well under 100 mg/day. That's the Upper Limit tied to neuropathy (Schaumburg 1983); a dose under ~25–50 mg is plenty outside a diagnosed deficiency.
- Prefer 5-MTHF folate. It's the one form choice with real PK data (Obeid 2020); the B12/B6 form choice matters less.
